The Russian, who has been off form as of late, defeated American Christopher Eubanks 6-4, 6-3.
Striking four aces, winning 89 percent of his first serve points and breaking serve on three occasions, Rublev will next face Nikoloz Basilashvili or No. 7 seed Arthur Rinderknech for a place in the semifinals.
Other results on the day saw Aleksandar Kovacevic, and Jesper de Jong advance to the second round.

Top seed Anna Kalinskaya reached the quarterfinals of the Singapore Tennis Open in Singapore on Wednesday, dismissing Simona Waltert from Switzerland 6-2, 7-6(3).
Needing one hour and 35 minutes to advance, Kalinskaya won 66 percent of her first serve points and broke serve on three occasions.
The 26-year-old Russian will next face Mananchaya Sawangkaew from Thailand for a place in the semifinals. Sawangkaew dismissed Cristina Bucsa from Spain 6-4, 6-4 in one hour and 39 minutes.
Other results on the day featured No. 6 seed Camila Osorio from Colombia dispatching Dominika Salkova 3-6, 6-2, 6-4, and Ann Li from the United States taking out Maria Timofeeva 6-3, 6-2.

No. 8 seed and recent Auckland winner Clara Tauson reached the quarterfinal stage of the Upper Austria Ladies Linz in Linz on Wednesday, defeating Sorana Cirstea 6-2, 6-4.
Needing 90 minutes to advance, Tauson struck one ace, won 71 percent of her first serve points and broke serve on four occasions.
The Danish star will next face either No. 2 seed Elina Svitolina or Anna Blinkova.
Other results on the day saw No. 7 seed Elina Avanesyan take out Katerina Siniakova 6-3, 6-2.
